I have a 2010 standard transmission and the shift knob is sort of loose. I can twist it clockwise and counter-clockwise fairly easily.
How can I tighten it up?

Use some thread-lock (blue Locktite). You can get it at Canadian Tire. Unscrew the know, put thread-lock on the threads and screw the know back on.

Seems like the leather is attached to it. Does it just come off?

I am assuming you mean the leather boot (it's really vinyly), not leather covering the shift knob itself.
I can't be 100% sure about the 2010's, as I have a 2008, but on it (and everyother car I have had with a stick shift) the knob just unscrews. It's probably dragging the boot along with it because of friction -- just hold the boot close to the top while unscrewing the knob until the knob is clear of the boot.
